Assessment of the biological effect of natural silicon when entering the body of experimental animals with drinking water. Part 1

Introduction. The biological effect of natural silicon (Si) and its hygienic regulation in drinking water still remain subjects of controversy in the scientific literature and in the practice of providing favourable conditions for water use by the population. In this regard, an assessment was made of the effect of natural silicon contained in drinking water in concentrations close to the maximum permissible concentration on the body of laboratory animals.
 Materials and methods. Drinking waters containing natural Si at MPC levels in water were studied, their effect was assessed in a three-month experiment on 80 white outbred male rats. Observations were made of the general condition, appearance of the animals, body weight dynamics, water consumption. 
 The morphological composition of whole blood was studied. Biochemical indices of the condition of internal organs, including the liver, were determined 
 in serum samples (the ALT and AST activity, the level of total protein and albumin), pancreas (α-amylase activity), kidneys (creatinine content), as well as lactate dehydrogenase activity as a nonspecific indicator of tissue damage.
 Results. In animals received 17±3.4 mg/L silicon with drinking water at a hardness of 2.25±0.47 mg-eq/L, a significant decrease in ALT activity was found on the 30th and 90th days of the experiment, AST – on 90th day, increase in the content of total protein and albumin in the serum on the 30th day of the experiment. Drinking water with silicon concentrations of 20.5±4.1 mg/L and hardness 3.55±0.76 mg-eq/L decreased ALT activity and serum creatinine concentration on the 30th day and increased amylase activity on the 90th day of the experiment. The animals had the highest water consumption and intake of silicon from drinking water over the first month of the experiment.
 Limitations. Limitations of the study are related to the short duration of observation and the small number of points for determining biochemical parameters over time.
 Conclusion. Consumption of drinking water containing natural silicon for 90 days led to changes in biochemical indices, largely indicating the positive effect of the element in concentrations of 17±3.4 mg/L and 20.5±4.1 mg/L on the body of laboratory animals.

Plasma lipid profile and sex hormone levels in rabbits under paraceta-mol-induced oxidative stress

Changes in the redox balance at different levels, which lead to the accumulation of toxic radicals against the background of a decrease in the content of redox-active compounds, i.e., a state of oxidative stress (OS), are considered the leading pathogenetic factor in pathological conditions. Processes of peroxide oxidation of biological substrates, especially cell membrane lipids and lipophilic compounds have both regulatory and pathological effects in the body of animals and humans. For now, it remains relevant to establish the regularities of interaction of metabolism during OS development. Among the large number of existing OS models, attention is drawn to those that can be reproduced in clinical practice, as a complication of treatment measures or an error in the medical strategy, due to a constant influence on peroxidation processes in the body. Important among such means is paracetamol (acetaminophen, APAP) – one of the most common antipyretic and analgesic drugs. In addition to the known hepato-, nephro- and neurotoxic effects of its long-term and/or excessive use, there are data on reproductive toxicity and disorders of lipid metabolism. Therefore, the aim of our work was to determine the lipid profile and the level of sex hormones in male rabbits during paracetamol-induced OS. The study was performed on male rabbits of the Khila breed. According to the principle of analogs, a control group (n = 12) was formed, which were kept on a standard diet, and an experimental group (n = 12), whose animals were simulated the state of OS by oral injection of a solution of paracetamol with food at a dose of 300 mg/kg of weight body once during 21 days. The following results were obtained by determining the dynamics of changes in biochemical indicators in the rabbit body during chronic injection of paracetamol: in the rabbits of the experimental group, after the injection of paracetamol, the level of diene conjugates was 63.0 % higher, and the content of thiobarbiturate-active products was 1.26 times higher (P ˂ 0.05). A decrease in the activity of the antioxidant defence system was also noted – the amount of reduced glutathione decreased by 35.4 %, and the activity of superoxide dismutase decreased by 25.6 % (P ˂ 0.05). The obtained changes were consistent with the comprehensive indicator of total antioxidant activity, which on the 21st day of the experiment was 34.1 % lower than the values of the control group (P ˂ 0.05). At the same time, the level of steroid hormones also changed during the study – the level of total testosterone gradually decreased (on the 21st day – by 22.8 %, and at the end of the experiment – by 30.9 %, P ˂ 0.05), while the level of 17β-estradiol had an upward trend and on the 21st day was higher than the control indicators by 10.7 % (P ˂ 0.05). Significant changes in lipid metabolism were found in male rabbits – the total cholesterol content on the 21st day of the experiment exceeded the data of control animals by 25.3 %, the level of triacylglycerols – by 42.3 %, and the content of low-density lipoprotein cholesterol also increased by 1.1 times (P ˂ 0.05), and the high-density lipoprotein cholesterol level, on the contrary, was reduced by 17.0 % (P ˂ 0.05). In general, the obtained results indicate the relationship between the development of OS with changes in lipid metabolism and the balance of sex hormones, which allows us to use the model of paracetamol-induced OS in rabbits for further research.

Effects of Auricularia auricula-judae (Bull.) Quél. polysaccharide acid hydrolysate on glucose metabolism in diabetic mice under oxidative stress

BackgroundOxidative stress can lead to uncontrolled glucose metabolism and, thus, diabetes. Auricularia auricula-judae (Bull.) Quél. polysaccharides possess biological activities, such as antioxidant and hypoglycemic effects, but their mechanism of their acid hydrolysates on oxidative stress-injured glucose metabolism disorders is unclear. PurposeUsing diabetic mice, we investigated the effect of the acid hydrolysate of polysaccharides from Auricularia auricula-judae (Bull.) Quél. on improving diabetes. Study design and MethodsThe structural information of sample polysaccharides was measured by high performance gel permeation chromatography, nuclear magnetic resolution, and high performance liquid chromatography. The diabetic model was established by intraperitoneal injection of streptozotocin. For eight consecutive weeks, the mice were orally administered sample polysaccharides (100, 200, and 300 mg/kg b.w. per day) for intervention. The improvement effect of the samples on diabetes was explored by detecting the changes in biochemical indicators in mice, and the underlying mechanism was studied by transcriptomic and metabolomic analysis. ResultsThe results showed that acid hydrolysate of Auricularia auricula-judae (Bull.) Quél. polysaccharides consisted mainly of mannose, xylose, glucuronic acid, and glucose; its weight-averaged molecular weight was 6.3842 × 104 Dalton, its number average molecular weight was 2.9594 × 104 Dalton; and the molecule contained α-Glc(1→4)-, β-Glc(1→3)-, and β-Man(1→4)-linked glycosidic bonds. A total of 100 mg/kg b.w. per day sample was the best intervention concentration. After eight weeks of intervention, the sample polysaccharides significantly reduced dynamic blood glucose and serum lipids, enhanced antioxidant enzyme activities, promoted glucagon like peptide-1 and insulin secretion, improved insulin sensitivity and alleviated insulin resistance in diabetic mice. Transcriptomic and metabolomic analyses showed that sample polysaccharides was able to ameliorate disorders of glucose metabolism by modulating gene expression such as glucokinase; and modulate the state of oxidative stress in mice in vivo by regulating the glutathione metabolism pathway. ConclusionAcid hydrolysate of Auricularia auricula-judae (Bull.) Quél. polysaccharides improved glucose metabolism disorders by slowing down the oxidative stress injury in mice, thereby alleviating diabetes. This study provided a basis for determining the underlying mechanism of the antidiabetic effect of Auricularia auricula-judae (Bull.) Quél. polysaccharides, which would significantly improve the deep development and application of these materials in diabetes control.

CORRECTION BY ANTIOXIDANTS OF OXIDATIVE ACTIVITY IN AN EXPERIMENTAL MODEL OF PARKINSON'S DISEASE

The advantage of the reserpine model in the development of parkinsonian syndrome in experimental animals was shown: symptoms appeared as early as 2 h after reserpine administration and persisted for up to 3 days. Injection of reserpine at a dose of 3 mg/kg, quite clearly influenced behaviour. Significant changes in biochemical indicators, markers of oxidative stress, accompanying disorders of motor functions of experimental animals were revealed. The possibility of correcting the pathology with the help of some agents of antioxidant nature has been shown. Despite the lack of clarity regarding the efficacy of succinate, tocopherol, the study showed that the use of goji berries significantly reduces the risk of Parkinson's disease.

Environmental impacts of chlorpyrifos: Transgenerational toxic effects on aquatic organisms cannot be ignored

Chlorpyrifos (CPF) has been extensively used in the world and frequently found in natural environments, might cause a range of environmental issues and pose a health risk to aquatic species. However, investigation of its toxic effects on offspring after parental exposure has been neglected, especially for aquatic organisms such as fish. In the current study, the effects of chronic CPF exposure (3 and 60 μg/L) on adult zebrafish (F0) was investigated to determine its influence on adult reproductive capacity and offspring (F1 and F2). The results showed the existence of CPF both in F0 ovaries and F1 embryos and larvae, indicating that CPF could be transferred directly from the F0 adult fish to F1 offspring. After 90 d exposure, we observed that F0 female fish showed increased proportion of perinucleolar oocyte in the ovaries, decreased proportion of mature oocyte, and decreased egg production, but not in F1 adult. The transcriptomic analysis revealed that the disruption of metabolism during oocyte maturation in the CPF treatment zebrafish might interfere with F0 oocytes development and quality and ultimately influence offspring survival. For the larvae, the parental CPF exposure distinctly inhibited heart rate at 72 and 120 hpf and increased the mortality of F1 but not F2 larvae. The changes of biochemical indicators confirmed a disturbance in the oxidative balance, induced inflammatory reaction and apoptosis in F1 larvae. Furthermore, the changing profiles of mRNA revealed by RNA-seq confirmed an increased susceptibility in F1 larvae and figured out potential disruptions of ROS metabolism, immune system, apoptosis, and metabolism pathways. Taken together, these results show that chronic CPF treatment can induce reproductive toxicity, and parental transfer of CPF occurs in fish, resulting in transgenerational alters in F1 generation survival and transcription that raising concerns on the ecological risk of CPF in the natural environment.

Наднормативні концентрації хлороформу та сульфату алюмінію у питній воді та їхній вплив на метаболічні процеси в організмі лабораторних тварини

Objective: determination of the nature and features of changes in biochemical indicators in the blood of animals under conditions of chronic exposure to different concentrations of chloroform and aluminum sulfate in drinking water in an isolated and combined state to substantiate the criteria for hygienic evaluation of their adverse effects on the body. Materials and methods: combined effect of chloroform and aluminum sulfate on biochemical indicators. Results: According to the results of biochemical studies, it should be noted that the most pronounced changes were observed in groups of animals that were exposed to CLF and Al in isolation at the level of 3 MPC and 5 MPC, and in groups of animals with combined exposure to the specified substances - CLF + Al at the level of 3 MPC аnd 5 MPC. A decrease in the content of AsT and an increase in the level of AlT in the above experimental groups of animals may indicate functional disorders of the liver, especially its enzymatic function. The indicated changes in the content of glucose and amylase in the blood as indicators of carbohydrate volume may indicate the activation of the processes of glycogenesis and glycolysis in the animal body, which may lead to functional disorders of the pancreas. Іt was established that the nature and severity of the effects depended on the nature of the influence of the studied factors (isolated or combined action), their MPC levels, and the time of exposure. Conclusions. The nature and features of changes in biochemical indicators in the body of experimental animals during a six-month chronic sanitary-toxicological experiment with isolated and combined effects of chloroform and aluminum sulfate in drinking water may indicate metabolic disorders in organs and changes in the intensity of regenerative processes at the cellular level in response to the adverse effects of toxicants. Such a tendency to imbalance indicators can be a kind of indicator of metabolic compensatory changes in the body under the influence of a complex of factors, depending on the current level and duration of action.

Observation of the efficacy of parathyroidectomy for secondary hyperparathyroidism in hemodialysis patients: a retrospective study

PurposeParathyroidectomy (PTX) is commonly performed as a treatment for secondary hyperparathyroidism (SHPT) in patients with end-stage renal disease (ESRD). We aimed to evaluate the efficacy of PTX in patients with SHPT who underwent hemodialysis.MethodsThis retrospective study analyzed the clinical treatment of 80 hemodialysis patients with SHPT who underwent either total PTX with forearm auto transplantation (TPTX + AT) or subtotal parathyroidectomy (SPTX). We compared the changes in biochemical indices before and after surgery as well as the attenuation of intact parathyroid hormone (iPTH) in the TPTX and SPTX groups. We also evaluated clinical symptoms and quality of life using the Visual Analog Scale (VAS) and the Short Form-36 Questionnaire (SF-36) before and at 3, 6, and 12 months after surgery.ResultsSerum iPTH and serum phosphorus levels decreased significantly after surgery in 80 patients with SHPT (P < 0.05). Within one month of surgery, there was a difference in iPTH levels between the TPTX + AT and SPTH groups, but there was no difference over time. Patients experienced significant improvement in their clinical symptoms of restless leg syndrome, skin itching, bone pain, and joint pain at 1 week post operation (P < 0.001). Quality of life significantly improved after surgery, as assessed by SF-36 scores (P < 0.05). Hypocalcemia was the most common postoperative complication, occurring in 35% of patients. Within the first 12 months post surgery, 5 patients had a recurrence.ConclusionPTX is effective in rapidly reducing iPTH levels, improving calcium and phosphorus metabolism disorders, and enhancing patients’ quality of life by safely and effectively relieving clinical symptoms.

Lupenone improves type 2 diabetic nephropathy by regulating NF-κB pathway-mediated inflammation and TGF-β1/Smad/CTGF-associated fibrosis

BackgroundType 2 diabetic nephropathy is a common diabetic complication and the main cause of death in patients with diabetes. Research has aimed to find an ideal drug with minimal side effects for treating this disease. Banana peel has been shown to be anti-diabetic, with lupenone isolated from banana peel exhibiting antidiabetic and anti-inflammatory activities; However, the effects of lupenone on type 2 diabetic nephropathy are largely unknown. PurposeThis study aimed to investigate the ameliorative effect of lupenone on type 2 diabetic nephropathy, and its mechanism from both anti-inflammatory and anti-fibrotic perspectives. MethodsSpontaneous type 2 diabetic nephropathy db/db mouse models were given three levels of lupenone (24 or 12 or 6 mg/kg/d) via intragastric administration for six weeks, and irbesartan treatment was used for the positive control group. We explored the effects and mechanism of lupenone action using enzyme-linked immunosorbent assay, automatic biochemical analyzer, hematoxylin-eosin and Masson staining, real time-PCR, and western blotting. Concurrently, a high-sugar and high-fat diet combined with a low-dose streptozotocin-induced type 2 diabetic nephropathy rat model was used for confirmatory research. ResultsLupenone administration maintained the fasting blood glucose; reduced glycosylated hemoglobin, insulin, and 24 h proteinuria levels; and markedly regulated changes in biochemical indicators associated with kidney injury in serum and urine (including 24 h proteinuria, micro-albumin, N-acetyl-β-d-glucosaminidase, α1-micro-globulin, creatinine, urea nitrogen, uric acid, total protein, and albumin) of type 2 diabetic nephropathy mice and rats. Hematoxylin-eosin and Masson staining as well as molecular biology tests revealed that inflammation and fibrosis are the two key processes affected by lupenone treatment. Lupenone protected type 2 diabetic nephropathy kidneys by regulating the NF-κB-mediated inflammatory response and TGF-β1/Smad/CTGF pathway-associated fibrosis. ConclusionLupenone has potential as an innovative drug for preventing and treating diabetic nephropathy. Additionally, it has great value for the utilization of banana peel resources.

Comparing steamed and wine-stewed Rehmanniae Radix in terms of Yin-nourishing effects via metabolomics and microbiome analysis.

Rehmanniae Radix Praeparata (RRP), the processed root of Rehmannia glutinosa, has been widely used to treat Yin deficiency syndrome in traditional Chinese medicine. RRP is available in two forms: processed by steaming with water (SRR) or processed by stewing with yellow rice wine (WRR). Previous work has documented chemical differences in the secondary metabolomes and glycomes of SRR and WRR. This study aimed to compare SRR and WRR in terms of Yin-nourishing effects via metabolomics and microbiome analysis. ICR mice were orally administered with thyroxine for 14d to induce Yin deficiency. Changes in biochemical indices and histopathology were detected. Serum metabolomics analysis and microbial 16S rRNA sequencing were performed to compare the therapeutic effects and mechanisms between SRR and WRR in treating thyroxine-induced Yin deficiency. Both SRR and WRR decreased serum T3, T4 and MDA levels, and increased SOD activity. SRR more effectively decreased serum Cr, and ameliorated kidney injury, while WRR showed better regulation on ratio of cAMP/cGMP and serum TSH, and relieved thyroid injury. Both SRR and WRR regulated tyrosine, glycerophospholipid, and linoleic acid metabolism and the citric acid cycle. Additionally, SRR regulated fatty acid metabolism, while WRR influenced alanine, aspartate and glutamate metabolism, and bile acid biosynthesis. SRR significantly enriched the genera Staphylococcus and Bifidobacterium in the gut microbiome, while WRR significantly enriched the genera Akkermansia, Bacteroides and Parabacteroides, and decreased the abundance of Lactobacillus. SRR displayed better protective effects on kidney, while WRR showed stronger effects on thyroid in thyroxine-induced Yin deficient mice. These differences might be due to different regulating effects of SRR and WRR on the metabolome and gut microbiota.

Peculiarities of toxic effects produced by aluminum oxide nano- and microparticles under multiple inhalation exposure

Introduction. Aluminum oxide nanoparticles (Al2O3 NPs) are widely used in nanotechnologies employed in various branches including chemical, food, and medical industry and perfume and cosmetics production. This high demand for Al2O3 NPs, given the wide-scale development of nanoindustries, can, in its turn, lead to ambient air pollution that creates public health risks under long-term exposure to it. Given that, it seems relevant to perform profound investigation with its focus on pathogenetic features of toxic effects produced by these nanoparticles and comparatively analyze them with effects produced by a micro-sized chemical analog under inhalation exposure to introduce more effective prevention.&#x0D; Materials and methods. We examined chemical properties of Al2O3, nano- and microparticles (MPs) in an experiment on Wistar rats, comparatively analyzed the results and described pathogenetic features of toxic effects produced by the examined particles under multiple inhalation exposure. &#x0D; Results. The examined samples were a nanomaterial judging by such parameters as particle size, shape, surface area, and total pore volume. They differed substantially from their micro-sized analog. Exposure to Al2O3 NPs causes more pronounced changes in the behaviour of rats relative to MPs. Under exposure to Al2O3 NPs, aluminum concentrations were statistically significantly by 1.62–55.2 times higher in the lungs, liver, brain and blood. The concentration of the examined elements was by 1.55–7.65 times higher in these organs as compared to exposure to the micro-sized particles. Exposure to Al2O3 NPs induced changes in biochemical indicators of negative effects against the control (exposure to micro-sized particles). We established higher activity of ALT, AST, AP, LDH, and elevated levels of direct bilirubin, GABA, glutamine acid, and MDA against the same indicators in the control group. Pathomorphological changes were identified in the lungs, brain, heart, and liver under exposure to Al2O3 NPs whereas exposure to the micro-sized analog induced such changes only in the lungs. Exposure to NPs induced more apparent changes in tissue structures in many organs. &#x0D; Limitations. The study involved only multiple inhalation exposure to Al2O3 NPs and MPs in an experiment on Wistar rats.&#x0D; Conclusion. Al2O3 NPs are more toxic than their micro-sized chemical analog; this is evidenced by a greater number of organs where bioaccumulation occurs, more apparent pathomorphological changes and pathological functional disorders. The study results should be considered when developing hygienic recommendations aimed at preventing and minimizing negative effects produced by Al2O3 NPs on human health.

Study of the chronic toxicity of the disinfectant drug Iodopovidone

The article reflects the results of preclinical studies of the chronic toxicity of the disinfectant drug Iodopovidone on non-linear laboratory white rats. Disinfectant Iodopovidon produced by LLC "Basalt" (Ukraine) is a polymer complex of iodine in its composition. The purpose of our research was to establish the chronic toxicity of the new promising disinfectant drug Iodopovedon. The experiment was conducted on the basis of the laboratory and clinic "Vetmedservis" of the Faculty of Veterinary Medicine of the National University of Life and Environmental Sciences of Ukraine on non-linear laboratory white rats in compliance with biotic norms in accordance with the Law of Ukraine "On the Protection of Animals from Cruelty" and the "European Convention on the Protection of Domestic Animals". 10 animals were involved in the experiment (5 animals - experimental and 5 animals - control), groups were formed according to the principle of analogues. Biochemical indicators of blood serum (total protein, albumins, globulins, glucose, urea, creatinine and activity of enzymes AsLT, AsAT), hematological indicators of blood and organ mass ratio (liver, kidneys, heart) were determined. Determinations were made in accordance with generally accepted methods. As a result of the study of biochemical indicators, a difference in the levels of total protein between the experimental and control groups was established within 2.1% due to a decrease in the level of the globulin fraction by 0.3% and a decrease in the level of the albumin fraction by 3.7% and a tendency to lower glucose levels by 28.0%. There was also a trend of an increase in AST activity by 7.3% and a decrease in ALT activity by 3.3%. The level of creatinine did not undergo pronounced changes against the background of an increase in the level of urea by 11.7%. When examining hematological indicators, a slight decrease in the number of lymphocytes by 6.6% and an increase in monocytes and granulocytes by 5.5 and 13.6% were found, respectively. The established changes in biochemical indicators of blood serum and hematological indicators of blood are insignificant and unlikely. The results of studies of the relative mass of internal organs - liver, kidneys and heart, to the final body mass of animals, indicate the absence of deviations. Therefore, the disinfectant preparation Iodopovidon does not cause pronounced changes in the critically important organs of the liver, kidneys and heart, does not cause pronounced changes in the morphological parameters of the blood of animals, the biochemical composition and activity of enzymes in the blood serum of white laboratory rats. Changes in Biochemical Indicators of the Blood of Patients With Fatal COVID-19

The study of the COVID-19 course is relevant due to the fact that this disease causes changes in biochemical indicators of blood in the human body. To analyze the age category of patients with COVID-19, distribution of patients by districts and regions in the Southern region, as well as associated disease factors in patients with COVID-19 with a fatal outcome. Research purpose: analysis of the age category of patients with COVID-19, distribution of patients by districts and regions of the Southern region, as well as concomitant disease factors in patients with fatal COVID-19. Research materials and methods: a descriptive study based on the analysis of electronic medical records of patients containing complete information about the course of the disease and data from standard laboratory and instrumental methods of examination in a hospital setting. Most of the patients with COVID-19 and pneumonia were admitted in Osh city or 65%, Karasuu district 14%, Uzgen district 7%, 4% Kara-Kulja district and 2% each Alai, Chon-Alai, Aravan districts and 2% each Batken and Jala-Abad regions. Conclusion: an increase in the absolute number of neutrophils (N), a decrease in the absolute number of lymphocytes (L) and, as a result, an increase in the N/L ratio index were markers of a more severe course of the disease.

Haemato-biochemical Profiling in Buffaloes in Relation to Metabolic Changes during Transition Period

Background: Transition period is a critical period in which most dairy animals undergo a period of negative energy balance (NEB), hypocalcemia, insulin resistance, hypoglycemia and susceptibility to infectious diseases. The critical changes in biochemical indicators are well reported in dairy cows whereas; in dairy buffaloes limited data related to alterations in hemato-biochemical changes have been reported so far. The current study aimed to evaluate the hemato-biochemical changes in buffaloes during transition period. Methods: A total number of 210 buffaloes aged between 3 to 14 years with parity ranged from 1st to 4th, divided in three groups (n=70; each) during transition period were used for the study. Three groups viz Group I (-30 days), Group II (0 days) and Group III (+30 days) were subjected for evaluation of hemato-biochemical parameters. Result: Our study showed significant increase (p less than 0.05) in WBC (103/µl), lymphocytes and granulocytes count at the time of calving. Significant decrease (p less than 0.05) in the level of RBC, Hb and HCT were seen in Group-I and II. In biochemical analysis, parameters like albumin, total protein, triglyceride and A:G ratio showed significant (p less than 0.05) decrease from Group-I to II. Calcium and Ca:P ratio decreased significantly (p less than 0.05) along with suppressed levels of sodium and potassium levels in Group-III. Thus, the present study showed significance of hemato-biochemical analysis during transition period for possibility of occurrence of production diseases.

Effect and mechanism of acupuncture on Alzheimer's disease: A review.

With the development trend of an aging society, Alzheimer's disease (AD) has become an urgent problem in the field of medicine worldwide. Cognitive impairment in AD patients leads to a decline in the ability to perform daily living and abnormalities in behavior and personality, causing abnormal psychiatric symptoms, which seriously affect the daily life of patients. Currently, mainly drug therapy is used for AD patients in the clinic, but a large proportion of patients will experience drug efficacy not working, and even some drugs bring severe sleep disorders. Acupuncture, with its unique concept and treatment method, has been validated through a large number of experiments and proved its reliability of acupuncture in the treatment of AD. Many advances have been made in the study of the neurobiological mechanisms of acupuncture in the treatment of AD, further demonstrating the good efficacy and unique advantages of acupuncture in the treatment of AD. This review first summarizes the pathogenesis of AD and then illustrates the research progress of acupuncture in the treatment of AD, which includes the effect of acupuncture on the changes of biochemical indicators in AD in vivo and the specific mechanism of action to exert the therapeutic effect. Changes in relevant indicators of AD similarly further validate the effectiveness of acupuncture treatment. The clinical and mechanistic studies of acupuncture in the treatment of AD are intensified to fit the need for social development. It is believed that acupuncture will achieve new achievements in the treatment of AD as research progresses.

Evaluating the Benefits of TACE Combined with Lenvatinib Plus PD-1 Inhibitor for Hepatocellular Carcinoma with Portal Vein Tumor Thrombus.

This study evaluated the efficacy and safety of transarterial chemoembolization (TACE) combined with lenvatinib plus programmed death (PD)-1 inhibitor (TACE-L-P) versus TACE combined with sorafenib plus PD-1 inhibitor (TACE-S-P) in the treatment of hepatocellular carcinoma (HCC) with portal vein tumor thrombus (PVTT). The clinical data of patients with HCC and PVTT treated with TACE-L-P or TACE-S-P from January 2018 to March 2022 were collected. The Response Evaluation Criteria in Solid Tumors (RECIST) version1.1 and modified RECIST (mRECIST) standard were used to evaluate the therapeutic effect. The progression-free survival (PFS) and overall survival (OS) of the two groups were compared. Blood samples were collected before and after treatment to detect the changes of biochemical indicators, and the adverse events (AEs) related to treatment were recorded. A total of 165 patients were included in the study, including 80 patients receiving TACE-L-P treatment and 85 patients receiving TACE-S-P. Patients in the TACE-L-P group had longer median OS (21.7months vs. 15.6months, P = 0.0027), longer median PFS (6.3months vs. 3.2months, P < 0.0001), higher objective response rate (41.25% vs. 30.59%, P = 0.008), and higher disease control rate (86.25% vs. 62.35%, P = 0.008) than those in the TACE-S-P group. Multivariate analysis of the TACE-L-P group showed that VP classification of PVTT, Child-Pugh grade, interleukin-17 (IL-17), vascular endothelial growth factor (VEGF), procalcitonin (PCT), and C-reactive protein (CRP) were independent factors significantly affecting patients' OS (P < 0.05). There was no significant difference in the incidence and severity of AEs between the two groups. TACE-L-P treatment can improve the survival of patients with HCC and PVTT with an acceptable safety, but higher inflammatory indicators will affect the therapeutic effect.

Relative comparison of chronic kidney disease-mineral and bone disorder rat models.

Objective: The aim of this study is to establish a suitable animal model of chronic kidney disease-mineral and bone disorder (CKD-MBD) by comparing CKD-MBD rat models induced by 5/6 Nx, AN, and UUO, accompanied by a low-calcium and high-phosphorus diet. Methods: Sprague‒Dawley rats were randomly divided into four groups: control group, 5/6 nephrectomy (5/6 Nx) group, Adriamycin nephropathy (AN) group, and unilateral ureteral obstruction (UUO) group. Serum biochemical indices were measured to evaluate renal function, mineral and bone metabolism, the severity of CKD-MBD, and the status of bone transformation. Hematoxylin-eosin staining (HE) and Masson's trichrome (Masson) staining were used for histopathological analysis of the kidney. Goldner's trichrome (Goldner) and tartrate-resistant acid phosphatase (TRAP) staining were utilized to observe bone mineralization and osteoclasts in the femur, respectively. Micro-CT images were applied to study the structure of the femur. The expression levels of osterix and cathepsin K in the femur were measured by immunohistochemistry (IHC) to confirm the status of bone transformation. Results: The levels of serum creatinine (Scr) and blood urea nitrogen (BUN) in the 5/6 Nx and AN group rats were significantly higher than those in the control rats, and this change was accompanied by marked changes in the levels of calcium (Ca), phosphate (Pi), intact parathyroid hormone (i-PTH), fibroblast growth factor 23 (FGF23), osteocalcin (OC), and cross-linked C-telopeptide of type 1 collagen (CTX-1); UUO group rats exhibited slight and inconsistent variations in the levels of Scr, BUN, Ca, Pi, i-PTH, FGF23, OC, and CTX-1 in serum. Histopathological analysis of the kidney showed that the UUO group rats suffered serious fibrosis and 5/6 Nx group rats exhibited severe focal calcification. Histopathological analysis of the femur showed that the AN group rats had minimal bone mineralization and that the 5/6 Nx group rats had overactive osteoclasts. Micro-CT revealed that the AN model had the most severe bone destruction and that the 5/6 Nx model had the least severe bone loss among the three models. The expression of cathepsin K in the femur was significantly increased in all models, while the expression of osterix in the femur was only significantly increased in the 5/6 Nx model. Conclusion: 5/6 Nx, AN, and UUO accompanied by a low-calcium and high-phosphorus diet successfully induced CKD-MBD in rats. The 5/6 NX model presented the progression of high-turnover bone disease, with consistency between biochemical indices in serum and histomorphometric analysis of the femur, and the AN and UUO models developed a severe deterioration in bone quantity and severe bone resorption; however, the changes in biochemical indices were subtle in the UUO model, and liver injury was obvious in the AN model.

Efficacy and Safety of TACE Combined with Regorafenib Plus PD-1 Inhibitor in the Treatment of Hepatocellular Carcinoma After Sorafenib Resistance.

To evaluate the efficacy and safety of TACE combined with regorafenib plus PD-1 inhibitor as a second-line therapy for hepatocellular carcinoma after sorafenib resistance. The clinical data of 76 patients with hepatocellular carcinoma who were drug-resistant to sorafenib from September 2018 to May 2022 in the tumor intervention department were collected. Among them, 35 patients used TACE combined with regorafenib plus PD-1 inhibitor (TACE-R-P) as second-line treatment, and the remaining 41 patients used TACE combined with regorafenib (TACE-R) as second-line treatment. The mRECIST (modified Response Evaluation Criteria in Solid Tumors) standard was used to evaluate the therapeutic effect. The progression-free survival (PFS) and overall survival (OS) of the two groups were compared. Blood samples were collected before and after treatment to detect the changes in biochemical indicators, and the adverse events (AEs) related to treatment were recorded. A total of 76 patients were included in the study, including 35 patients receiving TACE-R-P treatment and 41 patients receiving TACE-R treatment. Patients in the TACE-R-P group had longer median OS (19.7months vs 15.2months, HR:0.7716, 95% CI:0.4767-1.2490, P=0.03), longer median PFS (6.3months vs 3.8months, HR:0.6032, 95% CI:0.3727-0.9763, P=0.0029), higher objective response rate (37.14% vs 19.51%, P=0.001) and higher disease control rate (71.43% vs 48.78%, P=0.001) than those in the TACE-R group. Multivariate analysis showed that Child-Pugh grade (B/A; HR=1.283, 95% CI: 0.623-1.707, P=0.014), PVTT (Yes/No, HR=1.455, 95% CI: 0.977-2.038, P=0.018), extrahepatic metastasis (Yes/No, HR=1.766, 95% CI: 1.135-2.302, P=0.022) and treatment option (TACE-R/TACE-R-P, HR=1.930, 95% CI: 1.461-2.850, P=0.017) were independent prognostic factors for OS. There was no significant difference in the incidence and severity of AEs between the two groups. TACE-R-P treatment can be more effective than TACE-R treatment for HCC after sorafenib resistance and can be given priority as a second-line treatment for HCC.

Effects of Acute High-Temperature Stress on Physical Responses of Yellowfin Tuna (Thunnus albacares)

To understand the physiological reactions of juvenile yellowfin tuna (Thunnus albacares) under acute high-temperature stress, this study measured the changes in biochemical indexes of serum, liver, gill, and muscle of yellowfin tuna under acute high-temperature stress (HT, 34 °C) and a control group (28 °C) for 0 h and 6 h, 24 h and 48 h. The rising speed of water temperature in the HT group was 2 °C/h and the timing started when the temperature reached 34 °C. In the HT group, there was no significant difference between the four adjacent times in cortisol and lactic acid concentration. Serum triglyceride, cholesterol, and alkaline phosphatase concentration were significantly different from the four adjacent times. The superoxide dismutase (SOD) activity in the liver and gills increased at 6 h and 24 h, and the gills and liver had antioxidant reactions in a short time. The malondialdehyde (MDA) concentration in the gills changed significantly at 6 h, while that in the liver did not change significantly. The gills were more sensitive to temperature stress than the liver and muscle. Acute high-temperature stress affected yellowfin tuna’s antioxidant enzymes and metabolic indexes, resulting negative trend in physiological indexes, indicating that yellowfin tuna juveniles are susceptible to elevated temperature.

MONITORAMENTO FISIOLÓGICO E BIOQUÍMICO DOS ATLETAS DE DANÇA ESPORTIVA

ABSTRACT Introduction The dancesport is an emerging sports competition that integrates competitive sports and dance; college students have been using the training for aesthetic purposes in body delineation. However, the biochemical alterations are still underexplored. Objective Explore the physiological and biochemical changes caused by dancesport. Methods The volunteers were submitted to a two-week high-intensity dancesport protocol. Changes in physiological and biochemical indicators were monitored before and after the experiment. Results The athletes exhibited individual changes in biochemical indicators (P&lt;0.05), with emphasis on an increase in hemoglobin (from 152.3g/L to 169.2g/L). The physiological indices also changed significantly, with an average body fat reduction of over 5%. Conclusion Dancesport proved to help improve its practitioners’ physical and biochemical function. Adenovirus infections in dogs: diagnostic features

In the general pathology of dogs, viral diseases occupy a leading place, and infectious hepatitis, the causative agent of which is a virus of the Adenoviridae family, is of particular concern. Recently, the virus has spread to many countries around the world, which indicates changes in the properties of the pathogen. At the same time, the epizootic situation concerning infectious diseases, including adenoviruses, whose development is influenced by the mass keeping of dogs, has worsened in Ukraine. The purpose of this study is to supplement, clarify, and generalize data on epizootological features, morphological and biochemical parameters of blood and pathological and anatomical changes in type I canine adenovirus. The following research methods were used to conduct the research: epizootological analysis, clinical (determination of the general clinical condition of animals), pathological and anatomical (detection of macroscopic changes), haematological (morphological and biochemical parameters of blood) and statistical (processing of digital data to determine the probability of changes in indicators). According to the results of comprehensive studies, it was proved that dogs of different breeds, including mongrels, are susceptible to type I adenovirus, and the peak manifestation of the disease is the spring-summer period in animals aged one to two years. Infectious hepatitis is characterized by the development of erythrocytopenia, a decrease in haematocrit, leucocytosis, and lymphocytopenia. Changes in biochemical indicators are characterized by a decrease in haemoglobin, creatinine, urea, glucose in the blood, and a decrease in the activity of α-amylase and an increase in the content of total bilirubin, hyperfermentaemia of alkaline phosphatase and alanine aminotransferase. The most pronounced pathoanatomical changes are an increase in the size of the liver with the development of necrosis, and in the gastrointestinal tract-haemorrhages in the small intestine. The multisystem pathogenic effect of the virus is characterized by nephrosis, pinpoint haemorrhages in the pancreas, and inflammatory changes have been established in the lymph nodes. As a result of an experimental study of haematological parameters and pathoanatomical changes, a complex pathogenesis of the disease with multiple organ failure was established. In the study of infectious diseases of dogs, the most important aspect is clinical and diagnostic information content. Therefore, scientific research on a more profound understanding of the diagnosis of canine adenovirus will determine the algorithm for justifying the diagnosis
